What is CRISPR-Cas9?

CRISPR-Cas9 is a type II CRISPR-Cas system. The CRISPR-Cas9 system from Streptococcus pyogenesis a simple and versatile tool for RNA guided genome editing (RGE) in different organisms. During Cas9 mediated RGE, a single or duplex short RNA molecule (guide RNA or gRNA) directs Cas9 to target the desired DNA site for genome modification or transcriptional control. The gRNA-Cas9 complex recognizes the targeted DNA by gRNA-DNA pairing between 5’-end leading sequence of gRNA (referred as gRNA spacer) and one DNA strand (complementary stand of protospacer). Cas9 also requires the presence of protospacer-adjacent motif (PAM) in the target site following the gRNA-DNA pairing region. The CRISPR-Cas9 system can be reprogrammed to target any DNA site with a PAM by exchanging the approximate 20 nt long gRNA spacer sequence. But the gRNA-Cas9 complex also recognizes PAM sites that match the gRNA spacer imperfectly, resulting off-target risk in genome editing. With this website, we provide a resource of highly specific gRNA spacer sequences for seven different plant species.

What is CRISPR-PLANT

CRISPR-PLANT is a platform to help researchers to design and construct specific gRNAs for CRISPR-Cas9 mediated genome editing in plants. In this database, gRNA spacer sequences with specificity information are available for seven model plant species or crop species.
